NM_006254.4(PRKCD):c.1117G>C (p.Glu373Gln) was classified as Uncertain significance for Autoimmune lymphoproliferative syndrome, type III caused by mutation in PRKCD by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glutamic acid, which is acidic and polar, with glutamine, which is neutral and polar, at codon 373 of the PRKCD protein (p.Glu373Gln). This variant is present in population databases (rs782345985, gnomAD 0.006%). This variant has not been reported in the literature in individuals affected with PRKCD-related conditions. An algorithm developed to predict the effect of missense changes on protein structure and function outputs the following: PolyPhen-2: "Benign". The glutamine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
