This variant lies in the FLNC gene (transcript NM_001458.5) at the canonical splice donor site of the intron immediately after coding-DNA position 1549, where T is replaced by G; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: Variant summary: FLNC c.1549+2T>G is located in a canonical splice-site and is predicted to affect mRNA splicing resulting in a significantly altered protein due to either exon skipping, shortening, or inclusion of intronic material. Several computational tools predict a significant impact on normal splicing: Four predict the variant abolishes a canonical 5' splicing donor site. However, these predictions have yet to be confirmed by functional studies. The variant was absent in 248898 control chromosomes (gnomAD). To our knowledge, no occurrence of c.1549+2T>G in individuals affected with Cardiomyopathy and no experimental evidence demonstrating its impact on protein function have been reported. Three ClinVar submitter have assessed the variant since 2014, and all submitters classified the variant as likely pathogenic. Based on the evidence outlined above, the variant was classified as likely pathogenic.
