Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_017934.7(PHIP):c.5051A>T (p.Asp1684Val), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces aspartic acid, which is acidic and polar, with valine, which is neutral and non-polar, at codon 1684 of the PHIP protein (p.Asp1684Val). This variant is present in population databases (rs769265811, gnomAD 0.002%). This variant has not been reported in the literature in individuals affected with PHIP-related conditions. Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is not expected to disrupt PHIP protein function with a negative predictive value of 95%. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
