NM_017617.5(NOTCH1):c.3190G>A (p.Asp1064Asn) was classified as Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D1064N variant (also known as c.3190G>A), located in coding exon 20 of the NOTCH1 gene, results from a G to A substitution at nucleotide position 3190. The aspartic acid at codon 1064 is replaced by asparagine, an amino acid with highly similar properties. This variant has been detected in an individual with hypoplastic left heart (Stanley KJ et al. Eur J Hum Genet. 2024 Jul;32(7):795-803). This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
