This variant lies in the MYPN gene (transcript NM_032578.4) at coding-DNA position 3074, where A is replaced by G; at the protein level this means replaces glutamine at residue 1025 with arginine — a missense variant. Submitter rationale: Algorithms developed to predict the effect of missense changes on protein structure and function do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0"). This sequence change replaces glutamine with arginine at codon 1025 of the MYPN protein (p.Gln1025Arg).
