This variant lies in the RUNX1 gene (transcript NM_001754.5) at coding-DNA position 994, where G is replaced by C; at the protein level this means replaces aspartic acid at residue 332 with histidine — a missense variant. Submitter rationale: This sequence change replaces aspartic acid, which is acidic and polar, with histidine, which is basic and polar, at codon 332 of the RUNX1 protein (p.Asp332His). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with acute lymphoblastic leukemia (PMID: 34166225). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed for this missense variant.
