This variant lies in the TGFB3 gene (transcript NM_003239.5) at 5 bases into the intron immediately before coding-DNA position 755, where T is replaced by C. Submitter rationale: The c.755-5T>C intronic variant results from a T to C substitution 5 nucleotides upstream from coding exon 5 in the TGFB3 gene. This alteration has been reported in a sudden unexplained death case, and variants were also reported in other cardiac-related genes (Campuzano O et al. Forensic Sci Int, 2014 Dec;245:30-7). This nucleotide position is not well conserved in available vertebrate species. In silico splice site analysis predicts that this alteration will not have any significant effect on splicing. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
