Pathogenic for Charcot-Marie-Tooth Neuropathy X — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000023.11:g.(?_71223168)_(71224579_?)del, citing Invitae Variant Classification Sherloc (09022015): A gross deletion of the genomic region encompassing the full coding sequence of the GJB1 gene has been identified. The boundaries of this event are unknown as the deletion extends beyond the assayed region for this gene and therefore may encompass additional genes. Whole-gene deletions of GJB1 have been reported in the literature in individuals and to segregate in families affected with Charcot-Marie-Tooth disease (PMID: 11266688, 9760211, 10586261, 10587015, Invitae database). Loss-of-function variants in GJB1 are known to be pathogenic (PMID: 17353473). For these reasons, this variant has been classified as Pathogenic.
