Pathogenic for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_003242.6(TGFBR2):c.1058C>T (p.Ser353Phe), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the TGFBR2 gene (transcript NM_003242.6) at coding-DNA position 1058, where C is replaced by T; at the protein level this means replaces serine at residue 353 with phenylalanine — a missense variant. Submitter rationale: This sequence change replaces serine, which is neutral and polar, with phenylalanine, which is neutral and non-polar, at codon 353 of the TGFBR2 protein (p.Ser353Phe). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with clinical features of TGFBR2-related conditions (Invitae). In at least one individual the variant was observed to be de novo. ClinVar contains an entry for this variant (Variation ID: 477533). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt TGFBR2 protein function with a positive predictive value of 95%. For these reasons, this variant has been classified as Pathogenic.
