NM_001365276.2(TNXB):c.7168+11G>A was classified as Benign by Women's Health and Genetics/Laboratory Corporation of America, LabCorp, citing LabCorp Variant Classification Summary - May 2015: Variant summary: TNXB c.7168+11G>A alters a nucleotide located at a position not widely known to affect splicing. Consensus agreement among computation tools predict no significant impact on normal splicing. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 0.00051 in 242044 control chromosomes, predominantly at a frequency of 0.0066 within the East Asian subpopulation in the gnomAD database, including 1 homozygote. The observed variant frequency within East Asian control individuals in the gnomAD database exceeds the estimated maximal expected allele frequency for disease-causing variants in TNXB. To our knowledge, no occurrence of c.7168+11G>A in individuals affected with TNXB-related conditions and no experimental evidence demonstrating its impact on protein function have been reported. ClinVar contains an entry for this variant (Variation ID: 4773187). Based on the evidence outlined above, the variant was classified as benign.
