Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_177438.3(DICER1):c.739A>G (p.Thr247Ala), citing Ambry Variant Classification Scheme 2023: The p.T247A variant (also known as c.739A>G), located in coding exon 6 of the DICER1 gene, results from an A to G substitution at nucleotide position 739. The threonine at codon 247 is replaced by alanine, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
