Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_177438.3(DICER1):c.5169C>G (p.Asp1723Glu), citing Ambry Variant Classification Scheme 2023: The p.D1723E variant (also known as c.5169C>G), located in coding exon 23 of the DICER1 gene, results from a C to G substitution at nucleotide position 5169. The aspartic acid at codon 1723 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
