Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_177438.3(DICER1):c.5089A>G (p.Ile1697Val), citing Ambry Variant Classification Scheme 2023: The p.I1697V variant (also known as c.5089A>G), located in coding exon 22 of the DICER1 gene, results from an A to G substitution at nucleotide position 5089. The isoleucine at codon 1697 is replaced by valine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
