Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_177438.3(DICER1):c.2154G>C (p.Glu718Asp), citing Ambry Variant Classification Scheme 2023: The p.E718D variant (also known as c.2154G>C), located in coding exon 13 of the DICER1 gene, results from a G to C substitution at nucleotide position 2154. The glutamic acid at codon 718 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
