This variant lies in the DICER1 gene (transcript NM_177438.3) at coding-DNA position 1886, where C is replaced by T; at the protein level this means replaces threonine at residue 629 with methionine — a missense variant. Submitter rationale: To the best of our knowledge, the DICER1 c.1886C>T (p.T629M) variant has not been reported in individuals with DICER1-related disease. It was observed in 1/34588 chromosomes of the Latino subpopulation in the large and broad cohorts of the Genome Aggregation Database (http://gnomad.broadinstitute.org, PMID: 32461654). The variant has been reported in ClinVar (Variation ID 477072). Functional studies have not been performed, and in silico predictions of the variant's effect on protein function are inconclusive. The evidence is insufficient to meet ACMG/AMP criteria for classifying the variant as benign or pathogenic. Thus, the clinical significance of this variant is currently uncertain.
