Uncertain significance for Vitamin B2 deficiency — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_017986.4(SLC52A1):c.243_244del (p.Gln82fs), citing Invitae Variant Classification Sherloc (09022015): This sequence change creates a premature translational stop signal (p.Gln82Glyfs*47) in the SLC52A1 gene. It is expected to result in an absent or disrupted protein product. However, the current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in SLC52A1 cause disease. This variant is present in population databases (rs772259157, gnomAD 0.0009%). This variant has not been reported in the literature in individuals affected with SLC52A1-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
