While this particular variant has not been reported in the literature, loss-of-function variants in DICER1 are known to be pathogenic (PMID: 19556464, 21266384). This sequence change deletes 1 nucleotide from exon 9 of the DICER1 mRNA (c.1417delT), causing a frameshift at codon 473. This creates a premature translational stop signal (p.Tyr473Ilefs*7) and is expected to result in an absent or disrupted protein product.
