Uncertain significance for Charcot-Marie-Tooth disease type 4 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001370298.3(FGD4):c.1151C>T (p.Ala384Val), citing Invitae Variant Classification Sherloc (09022015): This variant has not been reported in the literature in individuals with an FGD4-related disease. This sequence change replaces alanine with valine at codon 247 of the FGD4 protein (p.Ala247Val). The alanine residue is highly conserved and there is a small physicochemical difference between alanine and valine. This variant is not present in population databases (ExAC no frequency). Algorithms developed to predict the effect of missense changes on protein structure and function do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C55"). In summary, this variant has uncertain impact on FGD4 function. The available evidence is currently insufficient to determine its role in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
