Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_003901.4(SGPL1):c.932C>G (p.Pro311Arg), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces proline, which is neutral and non-polar, with arginine, which is basic and polar, at codon 311 of the SGPL1 protein (p.Pro311Arg). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with congenital nephrotic syndrome (PMID: 35972040). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is expected to disrupt SGPL1 protein function with a positive predictive value of 80%. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
