This variant lies in the COL6A2 gene (transcript NM_001849.4) at coding-DNA position 2334, where C is replaced by A; at the protein level this means replaces aspartic acid at residue 778 with glutamic acid — a missense variant. Submitter rationale: Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain.
