Uncertain significance for Hypogonadotropic hypogonadism 1 with or without anosmia — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000216.4(ANOS1):c.2038T>C (p.Tyr680His), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the ANOS1 gene (transcript NM_000216.4) at coding-DNA position 2038, where T is replaced by C; at the protein level this means replaces tyrosine at residue 680 with histidine — a missense variant. Submitter rationale: This sequence change replaces tyrosine, which is neutral and polar, with histidine, which is basic and polar, at codon 680 of the ANOS1 protein (p.Tyr680His). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with ANOS1-related conditions. An algorithm developed to predict the effect of missense changes on protein structure and function outputs the following: PolyPhen-2: "Probably Damaging". The histidine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
