Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001128148.3(TFRC):c.313G>C (p.Glu105Gln), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glutamic acid, which is acidic and polar, with glutamine, which is neutral and polar, at codon 105 of the TFRC protein (p.Glu105Gln). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with TFRC-related conditions. An algorithm developed to predict the effect of missense changes on protein structure and function outputs the following: PolyPhen-2: "Benign". The glutamine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function.
