Uncertain significance — the classification assigned by Ambry Genetics to NM_001166108.2(PALLD):c.2635T>G (p.Cys879Gly), citing Ambry Variant Classification Scheme 2023: The p.C375G variant (also known as c.1123T>G), located in coding exon 6 of the PALLD gene, results from a T to G substitution at nucleotide position 1123. The cysteine at codon 375 is replaced by glycine, an amino acid with highly dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. The evidence for this gene-disease relationship is limited; therefore, the clinical significance of this alteration is unclear.
