Uncertain significance for Pyogenic arthritis-pyoderma gangrenosum-acne syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_003978.5(PSTPIP1):c.1172_1176del (p.Glu391fs), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the PSTPIP1 gene (transcript NM_003978.5) at coding-DNA position 1172 through coding-DNA position 1176, deleting 5 bases; at the protein level this means shifts the reading frame starting at glutamic acid residue 391, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: This sequence change is expected to alter the c-terminus of the PSTPIP1 protein (p.Glu391Glyfs*71). While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 26 amino acid(s) of the PSTPIP1 protein and extend the protein by 44 additional amino acid residues. This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with PSTPIP1-related conditions. Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
