Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_181783.4(TMTC3):c.1123G>A (p.Glu375Lys), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glutamic acid, which is acidic and polar, with lysine, which is basic and polar, at codon 375 of the TMTC3 protein (p.Glu375Lys). This variant is present in population databases (rs750602559, gnomAD 0.003%). This missense change has been observed in individual(s) with TMTC3-related conditons (PMID: 32973946). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
