Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_016562.4(TLR7):c.1888C>T (p.His630Tyr), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces histidine, which is basic and polar, with tyrosine, which is neutral and polar, at codon 630 of the TLR7 protein (p.His630Tyr). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with clinical features of TLR7-related conditions (PMID: 34413140). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated. Experimental studies are conflicting or provide insufficient evidence to determine the effect of this variant on TLR7 function (PMID: 34413140). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
