This variant lies in the COQ4 gene (transcript NM_016035.5) at coding-DNA position 32, where G is replaced by C; at the protein level this means replaces arginine at residue 11 with proline — a missense variant. Submitter rationale: This sequence change replaces arginine with proline at codon 11 of the COQ4 protein (p.Arg11Pro). The arginine residue is weakly conserved and there is a moderate physicochemical difference between arginine and proline. This variant is not present in population databases (ExAC no frequency) and has not been reported in the literature in individuals with a COQ4-related disease.
