This variant lies in the MUSK gene (transcript NM_005592.4) at coding-DNA position 535, where A is replaced by T; at the protein level this means replaces asparagine at residue 179 with tyrosine — a missense variant. Submitter rationale: This sequence change replaces asparagine with tyrosine at codon 179 of the MUSK protein (p.Asn179Tyr). The asparagine residue is highly conserved and there is a large physicochemical difference between asparagine and tyrosine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with MUSK-related conditions.
