Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000006.12:g.49605831del, citing Invitae Variant Classification Sherloc (09022015): This sequence change is expected to alter the c-terminus of the RHAG protein (p.Val405Serfs*58). While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 5 amino acid(s) of the RHAG protein and extend the protein by 52 additional amino acid residues. This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with RHAG-related conditions. Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
