Pathogenic for Epidermodysplasia verruciformis — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_152468.5(TMC8):c.988G>T (p.Glu330Ter), citing Invitae Variant Classification Sherloc (09022015): This sequence change creates a premature translational stop signal (p.Glu330*) in the TMC8 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in TMC8 are known to be pathogenic (PMID: 12426567, 22158547). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with TMC8-related conditions. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. For these reasons, this variant has been classified as Pathogenic.
