Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change. The variant allele was found at a frequency of 6.5e-05 in 247854 control chromosomes. This frequency is not significantly higher than estimated for disease-causing variants in COL12A1, allowing no conclusion about variant significance. To our knowledge, no occurrence of c.607C>A in individuals affected with COL12A1-related conditions and no experimental evidence demonstrating its impact on protein function have been reported.
