Uncertain significance for Malignant hyperthermia, susceptibility to, 5 — the classification assigned by Color Diagnostics, LLC DBA Color Health to NM_000069.3(CACNA1S):c.1619+1G>A, citing ACMG Guidelines, 2015. This variant lies in the CACNA1S gene (transcript NM_000069.3) at the canonical splice donor site of the intron immediately after coding-DNA position 1619, where G is replaced by A; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: This variant causes a G>A nucleotide substitution at the +1 position of intron 11 of the CACNA1S gene. Splice site prediction tools suggest that this variant may have a significant impact on RNA splicing. To our knowledge, RNA studies have not been reported for this variant. This variant has not been reported in individuals affected with CACNA1S-related disorders in the literature. This variant has been identified in 1/31388 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
