Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Color Diagnostics, LLC DBA Color Health to NM_000090.4(COL3A1):c.2198G>A (p.Gly733Asp), citing ACMG Guidelines, 2015: This missense variant replaces glycine with aspartic acid at codon 733 of the COL3A1 protein. Computational prediction indicates that this variant may have a neutral impact on protein structure and function. Functional studies have shown that this variant changes one of the conserved glycine residues within the Gly-Xaa-Yaa repeats of the triple helix domain (a.a. 168-1196) (PMID: 7695699, 8218237, 19344236). To our knowledge, this variant has not been reported in individuals affected with COL3A1-related disorders in the literature. This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
