Uncertain significance for Malignant hyperthermia, susceptibility to, 1 — the classification assigned by Color Diagnostics, LLC DBA Color Health to NM_000540.3(RYR1):c.9685+1G>A, citing ACMG Guidelines, 2015: This variant causes a G to A substitution at the +1 position of intron 65 in the RYR1 protein. To our knowledge, functional studies have not been reported for this variant. This variant has not been reported in individuals affected with RYR1-related disorders in the literature. This variant has been identified in 1/152248 chromosomes in the general population by the Genome Aggregation Database (gnomAD). Loss of RYR1 gene function due to haploinsufficiency is not an established disease mechanism for autosomal dominant malignant hyperthermia susceptibility. Therefore, this variant is classified as a Variant of Uncertain Significance.
