Uncertain significance for Malignant hyperthermia, susceptibility to, 5 — the classification assigned by Color Diagnostics, LLC DBA Color Health to NM_000069.3(CACNA1S):c.695-2A>T, citing ACMG Guidelines, 2015: This variant causes A to T substitution at the -12 position in intron 5 of the CACNA1S gene. Splice site prediction tools suggest that this variant may have a significant impact on RNA splicing. Although this prediction has not been confirmed in published RNA studies, this variant is expected to result in an absent or disrupted protein product. This variant has not been reported in individuals affected with CACNA1S-related disorders in the literature. This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). Loss of CACNA1S gene function due to haploinsufficiency is not an established disease mechanism for autosomal dominant malignant hyperthermia susceptibility. Therefore, this variant is classified as a Variant of Uncertain Significance.
