This variant lies in the LMNB2 gene (transcript NM_032737.4) at coding-DNA position 1256, where C is replaced by G; at the protein level this means replaces serine at residue 419 with tryptophan — a missense variant. Submitter rationale: This sequence change replaces serine with tryptophan at codon 419 of the LMNB2 protein (p.Ser419Trp). The serine residue is highly conserved and there is a large physicochemical difference between serine and tryptophan. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with an LMNB2-related disease. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain.
