Uncertain significance for Wilson disease — the classification assigned by Color Diagnostics, LLC DBA Color Health to NM_000053.4(ATP7B):c.2468A>G (p.Glu823Gly), citing ACMG Guidelines, 2015: This missense variant replaces glutamic acid with glycine at codon 823 of the ATP7B protein. Computational prediction suggests that this variant may have deleterious impact on protein structure and function. To our knowledge, functional studies have not been reported for this variant. This variant has been reported in an individual affected with neonatal hyperbilirubinemia (DOI: 10.21203/rs.3.rs-4845629/v1). This variant has been identified in 4/280952 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
