Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Color Diagnostics, LLC DBA Color Health to NM_000138.5(FBN1):c.862+1G>A, citing ACMG Guidelines, 2015. This variant lies in the FBN1 gene (transcript NM_000138.5) at the canonical splice donor site of the intron immediately after coding-DNA position 862, where G is replaced by A; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: This variant causes a G to A nucleotide substitution at the +1 position of intron 8 of the FBN1 gene. Splice site prediction tools suggest that this variant may have a significant impact on RNA splicing. Although this prediction has not been confirmed in published RNA studies, this variant is expected to result in an absent or disrupted protein product. This variant has not been reported in individuals affected with FBN1-related disorders in the literature. This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). Although there is suspicion that this variant may be associated with disease, additional studies are necessary to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
