Pathogenic — the classification assigned by Clinical Genomics Laboratory, Laboratory for Precision Diagnostics, University of Washington to GRCh38/hg38 11p15.5-15.4(chr11:198510-3400939)x3, citing ACMG/ClinGen CNV Guidelines, 2019. This is a single-copy gain (three copies) of the chr11:198510-3400939 region (~3.20 Mb) on cytogenetic band 11p15.5-15.4. Submitter rationale: Patient also had arr[GRCh38] 4p16.3(49556_3910769)x1. This duplication encompasses a region of 11p15.5 in which genes are regulated differently on the maternal copy transmitted from the egg compared to the paternal copy transmitted from the sperm. Silver-Russell syndrome is predicted if this duplication is maternally-derived, whereas Beckwith-Wiedemann syndrome is predicted if it is paternally-derived.
