Uncertain significance for Methylmalonic acidemia with homocystinuria, type cblX — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_005334.3(HCFC1):c.5210C>G (p.Ala1737Gly), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces alanine, which is neutral and non-polar, with glycine, which is neutral and non-polar, at codon 1737 of the HCFC1 protein (p.Ala1737Gly). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with HCFC1-related conditions. This missense change has been observed to be homozygous, hemizygous or homoplasmic in an individual who did not have the expected clinical features for that genetic result (internal data). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated.
