Uncertain significance for Polyglandular autoimmune syndrome, type 1 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000383.4(AIRE):c.1409T>G (p.Leu470Arg), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces leucine, which is neutral and non-polar, with arginine, which is basic and polar, at codon 470 of the AIRE protein (p.Leu470Arg). This variant is present in population databases (no rsID available, gnomAD 0.01%). This variant has not been reported in the literature in individuals affected with AIRE-related conditions. Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is expected to disrupt AIRE protein function with a positive predictive value of 95%. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
