Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001291088.2(WDR87):c.6664G>C (p.Gly2222Arg), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glycine, which is neutral and non-polar, with arginine, which is basic and polar, at codon 2183 of the WDR87 protein (p.Gly2183Arg). This variant is present in population databases (rs767174501, gnomAD 0.08%), and has an allele count higher than expected for a pathogenic variant. This variant has not been reported in the literature in individuals affected with WDR87-related conditions. An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
