This variant lies in the TGFBR2 gene (transcript NM_003242.6) at coding-DNA position 1547, where C is replaced by A; at the protein level this means replaces threonine at residue 516 with lysine — a missense variant. Submitter rationale: This sequence change replaces threonine, which is neutral and polar, with lysine, which is basic and polar, at codon 516 of the TGFBR2 protein (p.Thr516Lys). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with clinical features of Loeys-Dietz syndrome (PMID: 17979970). In at least one individual the variant was observed to be de novo. Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is expected to disrupt TGFBR2 protein function with a positive predictive value of 95%.
