The variant allele was found at a frequency of 0.00036 in 237782 control chromosomes, predominantly at a frequency of 0.0013 within the Latino subpopulation in the gnomAD database. The observed variant frequency within Latino control individuals in the gnomAD database exceeds the estimated maximal expected allele frequency for disease-causing variants in SCN11A. c.3506A>G has been observed in an individual affected with erythromelalgia like symptoms without clear evidence of causality (Kleggetveit_2016). These report(s) do not provide unequivocal conclusions about association of the variant with Familial episodic pain syndrome with predominantly lower limb involvement. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
