This variant lies in the DST gene (transcript NM_001723.7) at coding-DNA position 6838, where C is replaced by G; at the protein level this means replaces glutamine at residue 2280 with glutamic acid — a missense variant. Submitter rationale: This sequence change replaces glutamine with glutamic acid at codon 2280 of the DST protein (p.Gln2280Glu). The glutamine residue is weakly conserved and there is a small physicochemical difference between glutamine and glutamic acid. This variant is present in population databases (rs772001929, ExAC 0.004%). This variant has not been reported in the literature in individuals affected with DST-related conditions.
