Uncertain significance for Hereditary sensory neuropathy-deafness-dementia syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001130823.3(DNMT1):c.4148T>C (p.Val1383Ala), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces valine, which is neutral and non-polar, with alanine, which is neutral and non-polar, at codon 1383 of the DNMT1 protein (p.Val1383Ala). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with DNMT1-related conditions. Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is expected to disrupt DNMT1 protein function with a positive predictive value of 80%. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
