Uncertain significance for RASopathy — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_007373.4(SHOC2):c.806A>T (p.Gln269Leu), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glutamine, which is neutral and polar, with leucine, which is neutral and non-polar, at codon 269 of the SHOC2 protein (p.Gln269Leu). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with SHOC2-related conditions. Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed for this missense variant. However, the output from this modeling did not meet the statistical confidence thresholds required to predict the impact of this variant on SHOC2 protein function. This variant disrupts the p.Gln269 amino acid residue in SHOC2. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 35348676).
