Uncertain significance for Congenital disorder of glycosylation, type IAA — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_138459.5(NUS1):c.432T>G (p.Asn144Lys), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces asparagine, which is neutral and polar, with lysine, which is basic and polar, at codon 144 of the NUS1 protein (p.Asn144Lys). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with Parkinson disease (PMID: 30348779, 31582230). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
