Likely benign — the classification assigned by Laboratory for Molecular Medicine, Mass General Brigham Personalized Medicine to NC_000002.12:g.178565578_178565579delinsAA, citing LMM Criteria: Arg24284Cys in exon 275 of TTN: This variant results from a change affecting 2 b ases (either 72849C>T+72850C>T or 72849_72850delinsTT ? additional testing would clarify whether these are changes on the same of separate chromosomes). Both a re listed in dbSNP without frequency information (72849C>T, Phe24283Phe, rs18979 0119; 72850C>T, Arg24284Cys, rs185887755). This variant is not expected to have clinical significance due to a lack of conservation in mammals. Of note, gorilla and marmoset have a cysteine (Cys; this variant) at this position despite high nearby amino acid conservation. In addition, computational analyses (AlignGVGD, and SIFT) do not suggest a high likelihood of impact to the protein.
