However, this variant is not in the POLE exonuclease domain. While a complete picture of cancer risk associated with POLE is yet to be defined, many variants that are predicted to be deleterious but which are outside the POLE exonuclease domain have been identified that are none are associated with cancer risk. This variant does not segregate with colon polyps in one family that was evaluated. This evidence is consistent with a classification of likely benign. This analysis was performed in conjunction with the family studies as part of the University of Washington Find My Variant study.
